Project title: Mapping the dynamics of epigenomic and transcriptomic bases of cell differentiation in patients diagnosed with conjunctival MALT lymphoma by using SHARE-seq technology.
This project aims to conduct preliminary research in the field of transcriptome and epigenome analysis at the level of single cells in a group of patients diagnosed with conjunctival MALT lymphoma. The project provides for the study of the dynamics of the epigenomic and transcriptomic basis of cellular differentiation through the use of SHARE-seq technology, combining scRNA-seq and scATAC-seq technologies, which will allow to measure chromatin accessibility and gene expression at the level of single cells. The proposed SHARE-seq research methodology will allow to determine the full diversity of cell types and their states, regulatory elements that define them and the impact of common genetic variants on molecular processes in the cell types defined in the study. This is a highly scalable approach to measuring chromatin accessibility and gene expression in the same cell, demonstrating that changing chromatin accessibility to transcription factors affects the expression of specific genes. The obtained results will allow for a preliminary understanding of the complex ecosystem of cancer and immune cells in the conjunctival tumor and will be the basis for indicating their development along the pathways associated with specific genome aberrations.
